Zamburug'larga qarshi vosita, triazol hosilasi. Ta'sir mexanizmi zamburug'lar sitoxrom P450 tomonidan amalga oshiriladigan 14α-sterol demetilatsiyasini inhibe qilish bilan bog'liq, bu reaksiyaning zamburug'larning ergosterol sintezidagi asosiy bosqichi hisoblanadi.
In vitro vorikonazol keng spektrli zamburug'larga qarshi ta'sirga ega bo'lib, Candida spp. (shu jumladan, Candida krusei ftoroxazolga chidamli shtammlari va Candida glabrata va Candida albicans rezistent shtammlari) ga qarshi samarali hisoblanadi. Aspergillus spp. ning barcha o'rganilgan shtammlariga nisbatan ham fungitsid ta'sir ko'rsatadi. Shuningdek, Scedosporium va Fusarium kabi so'nggi paytlarda dolzarblashgan, zamburug'larga qarshi boshqa vositalarga kam sezgir bo'lgan patogen zamburug'larga qarshi ham faol.

Qisman yoki to'liq qarshi ta'sir ko'rsatilgan boshqa zamburug'li infektsiyalar quyidagilarni o'z ichiga oladi: Alternaria spp., Blastomyces dermatitidis, Blastoschizomyces capitatus, Cladosporium spp., Coccidioides immitis, Conidiobolus coronatus, Cryptococcus neoformans, Exserohilum rostratum, Exophiala spinifera, Fonsecaea pedrosoi, Madurella mycetomatis, Paecilomyces lilacinus, Penicillium spp. (jumladan, Penicillium marneffei), Philaphora richardsiae, Scopulariopsis brevicaulis va Trichosporon spp. (jumladan, Trichosporon beigelii).
In vitro sharoitida vorikonazolning Acremonium spp., Alternaria spp., Bipolaris spp., Cladophialophora spp., Histoplasma capsulatum klinik shtammlariga qarshi faolligi aniqlangan. Ko'pgina shtammlar 0.05 dan 2 mkg/ml gacha bo'lgan vorikonazol konsentratsiyasida o'sishni to'xtatadi.
In vitro Curvularia spp. va Sporothrix spp. ga qarshi faollik kuzatilgan, ammo uning klinik ahamiyati noma'lum.
